GENERAL INFORMATION Title: Chromium X controller upgrade Background Information: The Chromium X Controller has been a key tool in our collaborative research group for single-cell research. Utilizing Next GEM technology, it enables multi-dimensional single-cell analysis on a single system. This machine provides molecular readouts of cell activity across various dimensions, including gene expression, cell surface proteins, immune clonotypes, antigen specificity, and chromatin accessibility. Our lab has been using the previous model with 5' v2 kits, which remain compatible with the newest version, allowing us to continue using previously acquired kits. We use the 10X controller (and relevant reagents) for measuring the transcriptome of freshly obtained cells of the cerebrospinal fluid (CSF) and blood immune cells (peripheral blood mononuclear cells – PBMCs) from participants enrolled in clinical studies/trials to uncover biology. In addition, we have used the system for single-nuclei from autopsy tissue (mostly the brain). In future projects, we intend on using the 10X genomics system for the membranes coverin…
